Interviewer: Can you describe ViRexx Medical’s Chimigen™ therapeutic vaccine?


Dr. Rajan George: Chimigen therapeutic vaccine is used to produce immune responses in a host against infections which are difficult to produce immune responses, by targeting the vaccine to dendritic cells. The Chimigen platform can be extended to develop therapies for difficult-to-treat chronic infectious diseases.


Interviewer: Does that mean the Chimigen platform can be used to treat any infectious disease?


Dr. Rajan George: Yes, except in cases where the immune system is non-functional, as in the case of HIV. The Chimigen platform can be used to produce either a therapeutic vaccine or a prophylactic vaccine. This depends on the disease target and the antigen plugged into the platform. Some antigens have a use in treating infection, while others have a use in preventing an infection. Either one would be targeted to the dendritic cells. The therapeutic vaccine generates a cytotoxic T cell response. A prophylactic vaccine would generate a B cell response and antibody production.


Interviewer: From the way you’ve described the Chimigen™ vaccine, it appears potentially useful for many applications beyond Hepatitis B and C. How broad are the applications?


Dr. Rajan George: We should be able to use this platform for cancer therapy, depending upon the cancer antigen we use. We can plug in a specific cancer antigen into this platform, and the vaccine targeted to dendritic cells. The dendritic cells would process and present the right antigen, then generating immune responses (T &B cell) against the cancer. We are also evaluating some bioterrorist viruses, the biological weapons terrorists would use. We just started a project to look at one of those viruses to see if we can come up with the prophylactic vaccines to prevent diseases that would be caused by organism that could be used in bioterrorism.


Interviewer: Would the Chimigen™ vaccine be effective as a prophylactic against avian flu, H5N1?


Dr. Rajan George: It could work for bird flu if we just plugged in the bird flu antigen into the platform. Then we can use it as a prophylactic. It generates antibody to generate B-Cell response. You can produce a prophylactic vaccine using this platform. The Chimigen™ platform is quite adaptable.


Interviewer: How high is your confidence level in producing a prophylactic vaccine for the avian flu virus?


Dr. Rajan George: My thinking is that it is quite high. I think very highly of having a vaccine like that. But, the ultimate proof has to come from humans. Our HepaVaxx B clinical trial will give us a lot of information on how the technology really works. Until then, our optimism is based on laboratory results.


Interviewer: Can you describe what comprises the Chimigen platform?


Dr. Rajan George: The platform has two components. The first one is from the infectious agent. The second component is from a murine monoclonal antibody. Part one is fused with a fragment of part two by recombinant technology to produce a new entity, the Chimigen™ vaccine. We are recombining one thing with another. We have a virus which has certain antigens. We take one of those, and we produce a recombinant molecule with the fragment we have taken from a murine monoclonal antibody. Chimigen is the term we came up with to include the meaning of the full phrase, chimeric antigen. Chimeric means it comes from two different sources. We put them together and create a new molecule. One is from the virus. The other one is from the mouse, the monoclonal antibody. Now we have by recombinant methods produced a protein which is a chimeric protein. That’s the Chimigen™ vaccine.


Interviewer: How do you produce such a flexible vaccine, one that appears capable of treating nearly any infection?


Dr. Rajan George: To produce a Chimigen™ vaccine to treat nearly any infection, we start with an antigen (protein) from the infectious agent. We fuse it with a fragment called Fc of a mouse monoclonal antibody. This is done using recombinant methods. We end up with a new protein. This protein is made in a cell culture of commercially available insect cells. The protein is produced by the insect cells. From the culture, we purify this particular protein that we made. The insect cell system is just a tool. By virtue of its production in insect cells, the protein attains special properties which are useful in generating better immune responses. Producing this protein in insect cells gives it some very peculiar properties, which are different from our own mammalian proteins. Once we have it coming out of the cell, we purify it and make it really pure. Now we have a protein with the virus antigen murine monoclonal antibody with modified properties. This is a totally new entity.


Interviewer: What do you mean when you say, “useful in generating better immune responses”?


Dr. Rajan George: When a person has a chronic virus infection, his or her body ignores the virus and associated proteins. The body treats the virus as part of itself. The body does not recognize this virus as something foreign to it. Therefore the immune system does not attack the virus. But, by combining the virus antigen with a foreign protein such as the murine antibody fragment, the whole chimeric protein now is recognized by the body’s immune system as “foreign,” different from something of its own. In essence, this is a re-education of the immune system to switch its recognition of the virus from “self” to “foreign”.


Interviewer: From where did the scientific model come, and does it have similarities to another ViRexx Medical product, OvaRex MAb®?


Dr. Rajan George: This scientific model arose from discussions among the three lead scientists of the company, Dr. Tony Noujaim, Dr. Lorne Tyrrell, both founders of the company and myself. I am a biochemist by training and the collective thoughts of all of us went into the design of the Chimigen™ platform. One major similarity between Chimigen and OvaRex is that both involve a murine monoclonal antibody. Another similarity is that both target dendritic cells. The Chimigen model came from thoughts about targeting dendritic cells, but without the use of antibodies. OvaRex is a murine monoclonal antibody against the cancer antigen CA-125. The Chimigen™ vaccine has a fragment of a murine monoclonal antibody. OvaRex needs the CA-125 antigen in a cancer patient to bind to. The bound complex goes to the dendritic cells. The Chimigen™ vaccine does not need to look for the antigen in a patient because it already has the relevant antigen built in it.


Interviewer: It sounds as though the Chimigen™ vaccine acts in a similar way to OvaRex® in dealing with a hostile threat to the body’s health. How do they differ?


Dr. Rajan George: There are similarities and there are differences. OvaRex binds to the antigen CA-125. Then, the CA-125/Ovarex complex binds to the dendritic cells. The complex is internalized and processed. The peptides generated from the antigen are presented to the T cells, and the chain of events in the immune system gets stimulated. The activated cytotoxic T cells eliminate the cancer cells which contain the CA125 antigen. In the case of the Chimigen™ vaccine, the vaccine itself contains the antigen. It goes through the dendritic cell pathway and triggers the CTL response to clear the virus-infected cells. The system also produces antibodies to viral antigens, which bind virus and viral antigens and accelerate their removal. Because of the presence of the murine monoclonal antibody fragment, which is foreign to humans, along with the antigen from the virus, the body’s immune system treats this as a new threat and takes action. That is the immune response.


Interviewer: How would this work in treating Hepatitis B?


Dr. Rajan George: Developing a treatment for Hepatitis B chronic infection, for someone who already has the infection, would involve re-educating the immune system to react differently than it previously has. The infected person already has this virus and the derived antigens. If you put some more of the same antigens into the person, the person’s immune system is not going to know the difference His body is going to say, “Well, what’s the difference? I already have it. I am not going to do anything with it.” The body will ignore it. That’s what is called tolerance. With the Chimigen™ therapeutic vaccine, we have changed the body’s immune response to the virus.


Interviewer: How then have you changed the body’s response to the infection?


Dr. Rajan George: In a Hepatitis B chronic infection, let’s say I have the infection. My system is tolerating the virus. It’s ignoring the presence of the virus. While that is happening, the virus may be causing disease in with my liver. With time, it’s going to get my liver into trouble and my immune system has not responded adequately to remove the threat. We inject the protein – the one we just produced, which we call the Chimigen™ Therapeutic Vaccine – into the HBV chronic carrier, a person who has a chronic hepatitis B virus infection. What happens is when our protein is administered, the dendritic cells are going to look for anything new which enters the body. Those cells are the immune system’s first-line surveillance. The dendritic cells are going to see this new foreign protein, and they are going to think that this is different from what was previously inside. Their recognition of the molecule has changed from what it was before. Before the virus protein was recognized as a “self” protein. Now it is being recognized as a “foreign” protein. In chronic hepatitis B virus infection, the dendritic cells saw the virus as part of the “self” of the host, the vaccine changes the recognition of the virus protein as “foreign” to the host. Because the viral antigen is linked to the fragment of the mouse monoclonal antibody the direct the chimigen to dendritic cells it will enter the dendritic cell and be processed and stimulate an immune response.


Interviewer: And after the vaccine injection, what does the body see?


Dr. Rajan George: The body’s immune system see a new foreign antigen composed of a portion of the mouse monoclonal antibody linked to the viral antigen. It’s a foreign antigen.” The new “chimigen” stimulates an immune response to the antigen as well as the viral antigen. This is very important because the virus antigen was previously being ignored. Now, it’s being recognized as foreign through linked recognition of the mouse antigen as being foreign.


Interviewer: How do the dendritic cells react after they recognize this foreign threat?


Dr. Rajan George: The dendritic cells are the sentries of the immune system. They guard what comes in. When they recognize a “foreign situation,” what does the immune system do? It treats the whole molecule, the whole protein including the virus antigen, as foreign. The dendritic cells chop up this protein into small pieces called peptides. These peptides also are called “epitopes.” There are T cell epitopes which are smaller, and B cell epitopes which are longer. These small peptides bind to MHC I and activate Cytotoxic T lymphocytes (CTLs). The dendritic cells have a system where they put the T-cell epitope on another protein, MHC Class I, and bring it to the surface of the dendritic cell. They are presented as a complex on the surface of the dendritic cell to attract the T-cells. The T-cells come and see this, then get activated. Now, the activation is also specific to the virus protein. There are different varieties of T-cells, but the cytotoxic T-Cells are the most important in eliminating infections that already exist. The activated cytotoxic T-cells are the ones who do the attacking. They are the ones who start killing the virus infected cells.


Interviewer: And what about the B Cells?


Dr. Rajan George: That is the other side to this story. The dendritic cells can present another kind of peptide epitope. There is a second class of peptides, which are also produced when the protein is chopped up. The dendritic cells stimulate the B-Cells, B-Lymphocytes. And B-lymphocytes produce antibodies. The longer peptides bind to MHC II and activate B lymphocytes (B cells). B cells produce antibodies against the peptides. The antibodies are specific to the antigens we put in the Chimigen™ Therapeutic Vaccine. Antibodies bind to viral proteins that are on the surface of and block the ability of the virus to bind to a target cell to cause an infection and prevent the infection. This is the basis of a prophylactic vaccine. The antigen can bind to the invading virus and form a complex that the body eliminates. The B-Cells produce antibodies against the virus antigen, which we have put in the Chimigen™ vaccine. What do these antibodies do? The antibodies are specific to the antigen and bind to the viruses because they have the antigen. The system removes the virus by binding with the antibody. Also, the system removes infected cells using cytotoxic T-lymphocytes. Both of these actions are achieved by the Chimigen™ vaccine.


Interviewer: Aren’t there a lot of antibodies being investigated as therapeutics?


Dr. Rajan George: There are a lot of antibodies being investigated as therapeutics. OvaRex is the prime example. Avastin and Herceptin are two others, both of which are doing very well in the market. Another is Remicade, which is used to treat diseases such as rheumatoid arthritis. There are antibodies that are in various stages of clinical development, many are humanized antibodies where you want to avoid an immune response to the antibody. Our chimigen technology is new as we are trying to increase the immune response on a virus or a cancer through linked recognition. It is not found anywhere outside of our laboratories. This approach has not been tried before for chronic HBV or HCV infections.


Interviewer: Why would your vaccine work where others have tried and failed?


Dr. Rajan George: The reason is because of the novelty of the technology. We are re-educating the body’s own immune system to do the work by using the Chimigen™ technology. When you inject a xenotypic antibody , that is a non-human antibody that is linked to a specific antigen. The body recognizes the whole molecule as foreign and produces immune responses with both T - and B-Cell immunity. We believe that this enhanced immune response will be helpful in controlling the viral infection in the case of viral chimigens.


Interviewer: Much of the research has been within the laboratory. How much of this is hypothetical?


Dr. Rajan George: Our experiences so far have been mostly with isolated systems, meaning experimental systems outside of the body. For example, ViRexx’s Chimigen™ vaccine for treating chronic hepatitis B infection is what we call HepaVaxx B. This is waiting to go into Phase I clinical trials. We have done a lot of ex vivo experiments in the lab to evaluate the immune responses it can produce. We showed what we had predicted in theory has been true. We have also done some animal experiments, where the vaccine showed similar effects, again, as predicted. For HepaVaxx B, the animal results are also showing great progress and promise. We believe the Phase I studies will show safety and maybe some immunological data. The advanced clinical trials, Phase II and III, will tell us exactly what happens in humans (efficacy) with a chronic infection of Hepatitis B. I believe the Chimigen™ vaccine platform can make a difference in the area of immunotherapy of infectious diseases and cancer. For HCV, there is neither a therapeutic vaccine nor a prophylactic vaccine available in the market right now. Current available therapies are not very efficient, are expensive and have severe side effects. We do need more effective vaccines, both prophylactic and therapeutic, to prevent new hepatitis C virus infections and to eliminate existing infections. It is not an easy challenge but hepatitis C is an important target.

Nervous system training vs body part training

It is important to understand the two different aspects of training that is involved when you are trying to build bigger muscles. The two aspects are your nervous system versus your body part training. Both need to be incorporated in a holistic routine in order to maximize growth. In this article, we are going to demonstrate briefly some of the things that you need to consider.


The first thing to consider when examining your routine is the training effect on the nervous system. With the beginning trainee, their nervous system’s capacity to handle intense exercise is quite limited. Therefore, it makes sense that the individual should be working on their nervous system capacity as opposed to body part training. Unfortunately, most athletes or beginning trainees make the mistake of trying to copy a cookie cutter routine out of the magazines or a routine of a professional bodybuilder.


The problem with following a pro’s routine is that the routine generally far exceeds the capacity of the individual’s ability to train. The individual quickly spirals into overtraining, gets discouraged, and sometimes quits training altogether. Not an inspiring success story, that is for sure. Why is it that a novice trainee or even an intermediate bodybuilder over trains when following a pro’s routine? All things being equal it is because the advanced athlete or pro’s nervous system capacity for work is much higher than the aspiring trainees. The professional has probably spent 10 years minimum increasing his body’s capacity for training.


So, what does nervous system training involve? Well specifically with the beginner, almost any type of training will work. This fact is one of the biggest reasons there is so much confusion in training modalities in the body building market. Typically, any training that you perform at first places demands that are so much higher than your regular nervous system output, some results inevitably occur.


Unfortunately, most athletes have incorporated a great deal of bad habits in this early training, which ultimately spells failure down the road. For best results over the short and long term the beginner and intermediate trainee should incorporate training that increases the nervous system’s capacity for training. So what does this nervous system training include?


Nervous system training typically involves training specific exercises through specific planes and motions with an eye on frequency, volume, and exercise selection. When system training, one should train groups of body parts together to maximize the overload for a specific area. Now in order to create balanced growth as well as size, one must alternate back and forth between the nervous system training and body part training. This is so that a person can achieve maximal growth as well as minimizing any detriments to the physique. Always, you have body parts that are growing better than others and so the isolation body part training can allow you to re-evaluate your weak points and overcome these inherent weaknesses in your physique. By doing so, we build not only a large body but also a symmetrical body. The question then arises; how do I know when and where to switch between nervous system training and body part training?


There is no set rule on this however; one must look at it from this point; typically, the more inexperienced a trainee is, the more time they need to spend on nervous system training. Most trainees will benefit from two or three years of primarily nervous system training. So for example, one might spend three to six months just on nervous system training alone and briefly switch to a body part training for maybe 12 weeks before going back to another nervous system training routine. You would repeat this for a year or so, and then gradually working more body part training sessions in with the nervous system training.


Another thing to consider is during nervous system training, calorie restrictions is definitely not recommended. An overload of nutrients and calories is required in order to maximize growth. Often times during nervous system, we are not so much concerned with the finer aspects of physiques or physique building. Therefore, you pile in the calories to keep as much anabolic activity occurring so that the maximum amount of growth is achieved.


After several years of cycling between training styles one would benefit by making the switch to body part training and incorporate a low calorie diet. This factors a couple of things; one, you strip down the layer of body fat that has been built to reveal what is really muscle. Most people overestimate how much muscle they have and underestimate how much fat and water they have on their physique. At this stage in the athlete’s career it is beneficial that a low calorie regimen be followed at least for 12-20 weeks per year. By doing this frequently, it allows an individual to see the imbalances in physiques and correct them. The other aspect too is that it also helps the athlete to lower the metabolic step point; therefore, the individual in the next nervous system training will not have to consume as much food as he/she would if were continually staying on the nervous system training protocol.

Unfortunately, there is a lot of myths about the whole failure and intensity aspect of training. Article after article bombards the beginning and advanced trainee alike about the importance of training to failure. People are thought if they do not train to failure, they are not training intense enough or that they are some kind of wimp. Of course this type of mentality has nothing to do with science and as a matter of fact, will almost guarantee that the athlete reach a stagnantation point.


Optimally, the trainee never goes to failure and here is a couple of the reasons why:


First, when an individual is training naturally, the model of training is significantly different than the athlete who is using drugs. Most articles are written about pro-athletes or top amateurs who are using an incredible amount of drugs and therefore, really distort the science behind training. Because of this massive use of drugs, they endocrine systems operate completely different than that of the natural trainee and they are able to recover from mega high intensity training sessions.


Unfortunately over time, this endocrine system overload creates an eventual catabolic condition. If you check out any of the pros after their career, you can see how much muscle they actually lose. It is quite shocking. Often times, because of this lop-sided aspect of the endocrine system overload, the athlete’s career is stopped short due to disease. So once again, going back to the failure question; a person should train their exercises so that the last rep on every set is challenging, but it is not an all out effort. Now in order to get the intensity up volume is the key. So lots of sets for lots of reps is far more efficient and be far effective than short training sessions with max intensity. The scientific term of course is work out efficiency, but really the scientific term is not important.


The second aspect that needs to be addressed her is destroying the one hour workout myth. Once again, magazines and articles perpetuate this idea that the 45 minutes to one hour is all the time a person needs to build big muscles. This of course borders on ridiculousness. If one looks at, for example in the real world, an athlete such as an Olympic speed skater, these athletes perform incredibly high amounts of volume and incredible high amounts of frequency. Not only that, their time under tension far exceeds what most body builder’s workouts do. One other factor is speed skaters are performing an extensive amount of overload each day and very seldom does that athlete go to failure. Yet, look at the results. Speed skaters have some of the most largely developed legs in the entire world. Right there is a real world example that indicates that failure is not required and that training a few hours a week to achieve maximum development is a myth. I have yet to see any athlete that has trained a few hours and matched the legs of the world’s top speed skaters.


Once again here is a real world example that goes back to natural training versus drug usage. Not exceeding the endocrine system’s capacity to recover is critical in natural training. This is easier achieved if you train lots of sets and quite frequently. Bill Pearl, one of the greatest bodybuilding champions of all time, was a big believer in this principle and he reached a weight of 242 pounds at a height of 5’10”. Keep in mind that Bill Pearl never used any drugs to build his physique. An even more impressive consideration is that his physique was built in a time where the emphasis was not on the legs. In actuality, Bill probably had a 265-pound physique if you accounted for the lack of leg development that was in vogue during the late 60’s and early 70’s.


So once again for maximum results, a person needs to destroy the one-hour workout myth. Now, in the optimal situation, of course the individual would cycle between the two systems for maximal growth. So let us review the key points to get you on track and in the growth zone.


To maximize the growth, you need to go to nervous system training, high volume, exercise-specific techniques that involve body parts grouped together.


Body part training is going to be relatively between two to three times per week. Volume is going to be high and never train to failure. Southampton Castle


One of the chief ports of medieval England, Southampton preserves a wealth of medieval domestic architecture. Its flourishing Dark Age predecessor was abandoned in favor of the present site in the tenth century, and excavations have shown that this new town had earth and timber defenses from the beginning, no doubt as a defense against the Danes.


Over a mile in length, the walled circuit enclosed a roughly rectangular area. It had numerous bastions, mostly semi-circular, and larger towers ar the angles. Today, only the wall survives, along with parts of the north wall and a length near the southeast corner of the circuit. A tour of the wall may conveniently begin at the Bargate, the northern entrance to the old town and a very imposing one. The machicolated front is an early fifteenth century addition.


Behind it are twin half-round towers a century or so older, while the gate passage retains a Norman archway from an older structure. In contrast with the fortress-like outer face, the side facing the town has large windows lighting the story above the gate. This spacious chamber served as the guildhall in medieval times and later civic uses saved the gatehouse from demolition in later centuries. It was a major obstruction to traffic until the construction of a bypass in the 1930s, which relieved the problem but resulted in the destruction of the stretches of town wall on either side.


Beyond the Bargate, the wall leads west to the circular Arundel Tower, then soutward to the old quay. Shortly a kink in the circuit denotes the junction with the older castle wall. Southampton Castle was a royal stronghold first mentioned in the 1189s. Richard I and John rebuilt it in stone. The west curtain survives as the town wall, with a postern leading into a cellar from the castle's domestic buildings.

Tax haven raises 2006 entry price

While Monaco is a well known European tax haven, Andorra has remained little known outside of the financial community - despite enjoying the same tax advantages and arguably more private banking than her better known rival.

But Andorra has just raised property closing costs by introducing a three and a half per cent sale of goods and services tax on property purchases from January 1, 2006 - bringing the tax haven more in line with neighbouring France and Spain.


Demand for property in Andorra and Monaco is unlikely to be affected by the recent increases though, according to European tax haven specialists Tribune Properties.


‘Andorra and Monaco have historically seen an increase in property activity and residency applications when taxes are increasing elsewhere. The new German government has recently increased the top rate of income tax and the United Kingdom has seen an increase in the number of indirect taxes, making the zero per cent personal income tax both Andorra and Monaco offer an attractive preposition to high income earners.

But, most people in business don't make cars. In fact, they most likely sell clothes, books, time (through a job), commodity goods, intellect (through consulting), and so on. But, what particular business systems could possibly put into practice that will see their organisation become more proficient, easier to manage and produce better results for their customers?


A system can be defined as methods, organisation, or considered principles of procedure (Oxford Dictionary). And throughout your daily activities, there are certain tasks that you do every day or often enough that you have a certain way of doing them.


So, what types of activities could you systemize or create “considered principles of procedure” for, that will enable your organisation to be more efficient?


Emails - there are most likely a number of emails that arrive in your inbox daily or weekly, that you can create a system for processing them. Irrational fear chill out

While the whole world was masquerading in scary and outrageous costumes during Halloween, a team of scientists was making progress with the latest study on irrational fear. Results of this study are expected to expand current knowledge about treatments for major medical problems concerning irrational fear.


“We're taking all of what we learned from the basic studies of animals and bringing that into the clinical practices that help people. Things are starting to come together in a very important way,” said Stephen Maren, a psychology professor from the University of Michigan.


The National Institute of Mental Health revealed that about 40 million Americans suffer from anxiety disorders. Being a basic primal emotion, fear is a key to evolutionary survival which we share in common with animals. Aside from genetics, traumatic events are closely associated with the development of overwhelming and needless fear.


Ted Abel, a fear researcher at the University of Pennsylvania considers fear to be a funny thing. “ One needs enough of it, but not too much of it,” said Abel.


According to Armi Rowe, a Connecticut freelance writer and mother, she used to be one of those rational types who are usually calm under pressure. She is not afraid to downhill ski the treacherous black diamond trails of snowy mountains. One day, however, while coping with some serious illnesses in her family, she felt fear closing in on her while she was driving alone. She thought that the stab of pain on her chest was a heart attack and called 911. Later on, it was learned that what happened was an anxiety attack.


With the help of counseling, self-study coping exercises and introspection, Rowe learned to manage such attacks early on. David Carbonell, a Chicago psychologist specializing in treating anxiety disorders, said that there's a trick to panic attack. “You're experiencing this powerful discomfort but you're getting tricked into treating it like danger”, said Carbonell.


There is a possibility that by learning how fear runs through the brain and body, scientists can improve on how to dampen fear.


Amygdala, an almond-shaped part of the deep brain, is the fear hot spot. New York University psychology and neural science professor Elizabeth Phelps said that amygdala is not the one responsible for all fear response but acts like a burglar alarm that connects to everything else.


It was found that a certain chemical reaction in the amygdala is vital in the way mice and people learn to overcome fear, according to Emory University psychiatry and psychology professor Michael Davis. Mice never learn to counter their fears when that reaction is deactivated.


D-cycloserine, a drug already used to fight hard-to-treat tuberculosis, was found to strengthen that good chemical reaction in mice. When combined with therapy, it produces the same effect on people who have a fear of heights, and is now being tested in survivors of the World Trade Center attacks and the Iraq war.


Recent studies have shown that people easily recognize fear in other humans faster than other emotions. Other studies have suggested that bodily functions change just by being very afraid. One study found that very frightened people can withstand more pain than those not experiencing fear. Another found that experiencing fear or merely perceiving it in others improved people's attention and brain skills.


According to Carbonell, since the normal response for dealing with a real threat is either to flee or fight, the best way to deal with fear when the threat is unreal is to do just the opposite: “Wait it out and chill.”


Why an animated program needs voice actors

If you are producing an animated project, then you are going to need at least one voice actor, and you will have to know what to ask them about their skill sets. That means you need to go shopping for your voice actor—or actors in the plural, as the case may be-armed with a character list complete with a brief, one-sentence description of each character's voice requirements. Don't just hire a voice actor and hope you'll find out she can do what you need. Each voice actor is as different from every other, as each screen actor is different from his colleagues. After all, if the studio had cast Jack Nicholson as Vito Corleone, The Godfather would have been a different movie, even if Nicholson were Italian.


Whether you're casting an original character, a complex character with apparent contradictions in his nature—the rogue with a soft, gooey center, for instance—or an archetype like Gandalf of the Ring Trilogy and Dumbledore of the Harry Potter series, you need to know what a voice actor needs to be able to do in order to bring that character to life. You need to know what aspects of the voice are negotiable and which aren't.


A character's age or accent, for example, are generally not negotiable unless you are willing to change the very heart of your production. If you need a French accent, but find a voice actor who does British accents, but who is a genius at pulling off other difficult aspects of the character, it is perfectly all right to change the character to fit the actor—if you've found a gem of an actor. You have to make the call in the end, but you have to at least know what you want in the beginning, even if you find you are unable to get it.


You may even need special skills from a voice actor, like being able to do a child's voice. Chances are, you are not going to be able to find a child who is a voice actor, after all. But there are things you can do to create the illusion. Bart Simpson, for instance, is voiced by Nancy Cartwright.


All you really need when casting is a basic idea of the character's voice: gender, age, accent or lack thereof, and things like “smooth,” “rough,” “low,” “high,” “squeaky,” “sultry”--that sort of thing. And you need to know what kind of person he is, whether he's confident, sneaky, whatever. Then go shopping and see what happens.
